Oligo dT magnetic beads are magnetic microspheres covalently coated with oligodeoxythymine strands on their surface. Their core principle utilizes the base complementary pairing rule, where the Oligo dT strands specifically hybridize and bind to the PolyA tail, a characteristic feature at the 3' end of eukaryotic messenger RNA (mRNA), enabling the rapid and high-purity separation of mRNA from total RNA.
Simply put, Oligo dT magnetic beads act like a "specialized magnetic comb," precisely "extracting" mRNA with a PolyA tail from the RNA mixture.
