        • Cre-recombinase is an enzyme that recognizes specific DNA sequences called loxP sites. loxP sites are specific 34-base pair DNA sequences that serve as recognition and binding sites for Cre-recombinase. A typical loxP site consists of two 13-bp inverted repeats flanking an 8-bp spacer region. The orientation and location of loxP sites determine the outcome of the recombination event (e.g., deletion, inversion, or translocation of the DNA between the sites).

          The enzyme catalyzes the recombination between these sites, leading to the excision or inversion of the DNA between them, depending on their orientation. This allows for precise genome engineering by enabling the removal or rearrangement of specific DNA segments.

          This product CMV-LoxP-DsRed-LoxP-GFP-PGK-PuroR lentivirus is designed for Cre-recombinase effectiveness test. Here is the lentivirus format: DsRed followed by stop codon is bracketed with two LoxP sites before GFP open reading frame in this product. Transduce the target cells in vitro or in vivo with the lentivirus. DsRed signal will be available in the cells transduced with this virus in the absence of Cre-reccombinase expression. Fluorescence signal will switch from DsRed to GFP when cre-recombinase is expressed in the same target cells. It is convenient to monitor the Cre-Recombinase activity in the cells by visualizing the fluorescence signals.
