Klenow (3'→5'exo-) DNA polymerase is a proteolytic product of E. coli DNA polymerase I. It retains the DNA polymerase activity of DNA polymerase I but loses the 5'→3' exonuclease activity. The enzyme further removes the 3'→5' exonuclease activity through mutation. It can be used for DNA labeling by random primer method, DNA sequencing by dideoxy method, etc.
