To avoid the biosafety risks associated with traditional H7N9 virus neutralization assays that involve live virus, a pseudovirus has been developed based on the HIV lentiviral backbone, with the HA and NA proteins of H7N9 virus (Strain: A/Shanghai/4664T/2013; GenBank ID: AGI60292.1) embedded on its surface. This pseudovirus simulates the process of the live virus binding to receptors and entering receptor cells. Additionally, the pseudovirus carries GFP (green fluorescent protein) and Luciferase (firefly luciferase) reporter genes, allowing the evaluation of cell infection activity through fluorescence observation or luciferase activity detection. It can also be used to assess the ability of neutralizing antibodies to block pseudovirus infection of cells. The pseudovirus lacks self-replicating ability, ensuring high safety. It serves as an important tool for H7N9-related drug screening, neutralizing antibody activity detection, and vaccine efficacy evaluation.
