Fetal umbilical cords taken from natural delivery at full term were cryopreserved in P3 generation. Immunofluorescence assay showed that the cells expressed multiple mesenchymal stem cell specific markers and had good proliferation and differentiation potential. The test results for CD29, CD44, CD73, CD90, CD105, and CD166 were positive, and they could be induced to differentiate into osteoblasts, adipocytes, and chondrocytes. Using a complete culture medium of mesenchymal stem cells, the cells could increase by more than 15 times.
Medium and culture conditions: MSCM mesenchymal stem cell culture medium.
Gas phase: Air, 95%; Carbon dioxide, 5%.
Temperature: 37°C.
Freeze storage conditions: Basic medium+5% DMSO+20% FBS.
