This kit can directionally clone PCR products into any position of any vector at a concentration as low as 5 ng/μL, and can achieve sequential splicing cloning of up to 6 fragments at a time. Linearize the vector, and introduce 15-25 bp homologous sequences at the end of the linearized vector at the 5' ends of the forward and reverse PCR primers of the inserted fragment, so that the 5' and 3' ends of the inserted fragment PCR product contain the same as the linearized vector respectively. Completely identical sequences corresponding to both ends. The PCR product and linearized vector can complete the recombination reaction under the action of recombinase in only 50°C and 5-15 minutes.
