Background
EZ Cap Reagent AG (3' OMe) is a co-transcriptional capping reagent for in vitro transcription of 5’ capped mRNA resulting in a Cap 1 structure. The Cap reagent has shown to provide up to 98% capping efficiency in literature.
EZ Cap AG (3' OMe) contains the 5' N7-Methyl-3'-O-Methylguanosine similarly to mRNA capped using ARCA, which is different from EZ Cap AG with a 5' N7-Methylguanosine structure.
EZ Cap AG requires an AG initiator. EZ Cap AG could typically provide >90% capped material with a naturally occurring Cap 1 structure. EZ Cap is compatible with wild-type T7 polymerase. 5 mM EZ Cap and rNTPs is recommended. Template design is an integral part of any transcription. EZ Cap reagent AG 3’OMe should be used with a specific initiating sequence 5’ AG 3’.
Final capping efficiency is dependent on the EZ Cap Reagent, DNA template and final mRNA sequence. Secondary structure due to RNA length and base composition could affect final capping efficiency.
