Type III-A CRISPR-associated protein Csm6 is obtained from Enterococcus italicus. EiCsm6 is a dimeric RNA endonuclease that consists of a CARF domain which binds cyclic oligoadenylate and a HEPN domain with ribonuclease activity. EiCsm6 is activated by cyclic oligoadenylate second messengersby binding to the CARF domain, which causes the HEPN domain of EiCsm6 to be activated for RNA degradation. The CARF domain then degrades the cyclic adenylate activator (cA6) which acts as a switch to regulate CRISPR enzyme activity.
