Azido-PEG4-acid is a PEG analogue containing an azide group and is used as a PROTAC linker for the synthesis of Rucaparib-based PARP PROTAC degraders. With a terminal carboxylic acid, it is also a non-cleavable 4 unit PEG ADC linker used in the synthesis of antibody-drug conjugates (ADCs). The hydrophilic PEG spacer increases solubility in aqueous media. The azide group can react with alkyne, BCN, DBCO via Click Chemistry to yield a stable triazole linkage. The terminal carboxylic acid can be reacted with primary amine groups in the presence of activators (e.g. EDC, or DCC) to form a stable amide bond.
