Aminoacyl-tRNA Synthetase

Product: Aminoacyl-tRNA Synthetase, suspended in 10 mM Tris buffer (pH 7.6) containing 50% glycerol, 10 mM MgCl2, 10 mM KCl and 30 mM 2- mercaptoethanol.
Activity: 1-5 U/μl.

Application: This enzyme is used to catalyze the aminoacetylation of tRNAs with its correspondent amino acid.
Testing indicates that this enzyme can catalyze the aminoacetylation of lysine, proline and tryptophan with their corresponding tRNA. Aminoacetylation of other amino acid-tRNAs should be tested. Unit definition: One unit will activate and attach 0.1 picomole of H3- labeled lysine amino acid to tRNA in 30 minutes at 37°C. The activity of this enzyme is about 15 times higher then the one offered by our competitors: 5 units of this enzyme can suffice for a 60μl reaction volume. From bovine liver.

Background

RNA is most widely used as the primer of retroviral reverse transcriptases. The 3’ end of the transfer RNA binds to the complementary binding site on the viral RNA therefore initiating reverse transcription.

tRNA Lysine 3 is the primer of HIV-I, HIV-II, SIV and FIV.
tRNA Lysine 1,2 are the primers of BLV, HTLV-I and II, SSV, FLV, VV, CAEV, MPMV, SMR and EIAV.
tRNA Tryptophan and tRNA Proline are the primers of ASNV, SSV, FLV, BLV, MLV, HTLV-I and II.
Since it has been well documented that artificial transfer RNA does not posses the same biological properties as natural tRNA, human purified transfer RNAs are largely in demand for retroviral research, especially HIV related research.

The extraction and purification of transfer RNA from a natural source such as human placenta is a highly complex and delicate procedure.
