Amino-dPEG®4-t-boc hydrazide, product number 10041, is a useful crosslinking reagent. The free amine reacts with carboxylic acids and their active esters to form an amide bond. The boc-protected hydrazide can be uncovered using formic or trifluoroacetic acid and then reacted with a carboxylic acid or active ester to form a hydrazone. Hydrazone bonds are acid labile, thus allowing the conjugate or supramolecular construct to degrade under acidic conditions such as occur in lysosomes or the acidic extracellular microenvironments at sites of inflammation or tumor metastasis. The single molecular weight, discrete PEG (dPEG®) crossbridge is water-soluble and non-immunogenic, and it adds flexibility and increases the hydrodynamic volume of conjugates and supramolecular constructs that incorporate it.
Potential applications for Amino-dPEG®4-t-boc hydrazide include dendrimer construction, side-chain modification of amino acids, and antibody-drug conjugates. This product could also act as a building block for releasable labels such as biotin or dye or releasable payloads such as a small molecule drug or peptide.
