AAV9 Capsid Libraries are designed for the evolution and development of novel AAV capsids with specific characteristics based on AAV9 capsid. AAV9 Capsid Libraries have 7 to 12 amino acid random peptides insertion on the surface of viral capsid at VR-IV or VR-VIII region. AAV9 CAP gene with random peptide are driven by a CAG/P40 hybrid promoter. P40 promoter was used to drive CAP expression during AAV production. CAG promoter is used to express CAP in targeting cells or tissues.

Background
The AAV capsid libraries are random peptide insert library build on our ATHENA-II platform. They are used to evolute novel AAV capsids for different purposes. Each library has high library complexity which provides a huge number of screening resources for AAV capsid directed evolution study. With our novel library construction strategies, we offer generation of high complexity, random peptide insertion AAV capsid libraries at any site and at any peptide length in addition to the libraries listed in the catalog.In the premade AAV Capsid library, CAP gene with random peptide are driven by a CAG/P40 hybrid promoter. P40 promoter was used to drive CAP expression during AAV production. CAG promoter is used to express CAP in targeting cells or tissues. In the premade AAV capsid libraries, each library has complexity over 1e9 and has titer over 1e13 GC/ml. AAV viral libraries were produced at 200 copies/cell conditions in HEK 293T cells and purified with two rounds of CsCl ultracentrifuge.
