The sphere culture does not need a scaffold, and forms a cell sphere through the characteristics of adherent cells tending to gather, which is one of the most common and universal non-scaffold methods used for 3D cell culture. The commonly used spheres include embryonic bodies, breast spheres, tumor spheres, hepatocyte spheres and nerve spheres, which are used for CSC research. This medium does not contain serum, and is composed of recombinant stem cell growth protein, vitamins, minerals, etc., and small molecular compounds such as AMP, BMP inhibitor, Rock and SMOC-2 are added to inhibit the growth of miscellaneous cells and promote tumor formation of tumor stem cells.
