Nectin-4 (PVRL4) is a type I transmembrane glycoprotein that belongs to the nectin family of Ig superfamily proteins. Nectin-4 is expressed in the placenta, especially in endothelial cells. Nectin-4 ECD contains three Ig-like domains, including one N-terminal V-type and two C2-type. As cell adhesion molecules, Nectin-4 plays an important role in many biological processes during human development as well as adult life. Studies also showed that Nectin-4 acts as tumor-associated antigen in some of the breast, ovarian and non-small cell lung carcinomas, especially on those tumors with bad prognosis.
