ErbB2, also known as human epidermal growth factor receptor 2 (HER2), is a type I membrane glycoprotein that belongs to the epidermal growth factor (EGF) receptor family. ErbB2 is expressed in various tumor tissue including primary breast tumors and tumors from small bowel, esophagus, kidney and mouth. ErbB2 plays a key role in development, cell proliferation and differentiation. ErbB2 is reported to be an orphan receptor, with none of the EGF family of ligands able to activate it. Around 30% of breast cancers overexpress ErbB2, which is associated with increased disease recurrence and worse prognosis.
