Nebula Digital PCR System

DPBio Nebula Auto Digital PCR System is based on microfluidic technology that distributes nucleic acid samples and amplification systems into a large number of individual microdroplets. Each unit contains one to multiple nucleic acid target molecules to be examined, and the entire dispensing process conforms to the Poisson distribution. In each microreaction unit, after the target molecule is amplified by PCR, the instrument detects each microdroplet one by one. The copy number of the target molecule can be calculated by the Poisson distribution formula based on the number of negative or positive microdroplets and the total number of droplets.

Workflow
· Generation of reaction mixtures
The DPBio Digital PCR Kit and samples required for the experiment are mixed well in certain ratios.
· Microdroplet generation
The reaction mixture is transferred to the droplet generator chip and droplets are generated by the droplet generator. The chip uses a patented air pressure insensitive droplet generation technology, which can complete droplet generation for 32 samples in 3.5 min. The generated droplets are automatically collected without manual transfer, reducing droplet loss.
· PCR Amplification
DNA or RNA fragments within each microdroplet reaction unit are amplified by PCR to form large DNA clusters.
· Read / analyze microdroplet results
The amplified sample is transferred to the Digital PCR Reader. During the process, the sample is automatically transferred to the reading chip, and this closed experiment design can eliminate contamination. The Reader is available in a variety of settings, with 2-channel, 4-sample and 3- to 6-channel, 8-sample versions, and reads signals in about ten minutes or less.
