Self-renewal of colonic epithelial cells is driven by proliferation of stem cells and their progenitor cells located in the crypts, allowing human colonoid organs to display all the characteristics of human colonoid organs. The colonic epithelium is highly influential in terms of structure, cell type composition and self-renewal kinetics. The prospects for the study of human colon development and disease are unprecedented, and human colonoid organs may also have applications to the in vitro expansion of the colonic epithelium in regenerative biology.
