PEG aldehydes are mostly amine reactive pegylation reagents. They react with free amines, peptides and proteins forming an imine bond, which can be reduced afterwards or insitu by hydrides to form a stable secondary amine bond. However, with hydrazide substrates, hydrolytically unstable hydrazone bonds are formed, thus making reversible pegylation possible.
