The CCK-8 kit contains the compound WST-8 (chemical name: 2-(2-methoxy-4-nitrophenyl)-3-(4-nitrophenyl)-5-(2,4-disulfonatophenyl)-2H-tetrazolium monosodium salt) and is used for cell proliferation detection. Its mechanism of action is similar to MTT, where it is reduced by certain dehydrogenases in the mitochondria of cells in the presence of electron-coupling reagents, resulting in the formation of a highly water-soluble orange-yellow formazan. The faster the cell proliferation, the more formazan is generated, resulting in a deeper color. The absorbance at 450 nm is measured using a microplate reader, which can indirectly reflect the number of viable cells. The CCK-8 kit has been widely applied in the detection of bioactive factors, large-scale anti-tumor drug screening, and cell proliferation assays.
Compared to MTT and similar assays, the WST-8-based cell proliferation detection kit has lower cytotoxicity, higher sensitivity, and simpler operation.
