      • Overview
        • Bis-dPEG®25-NHS ester, product number 10968, is a monodisperse, homobifunctional, amine-reactive polyethylene glycol (PEG) crosslinker. Both ends of the dPEG® crosslinker terminate with N-hydroxysuccinimidyl (NHS) esters. NHS esters are presently the most widely used amine-reactive acylating reagents. This product crosslinks peptides and proteins with free amines such as the side chains of lysine. Consequently, Bis-dPEG®25-NHS ester is useful in proteomics research. Bis-dPEG®25-NHS ester increases the water solubility and hydrodynamic volume of conjugates made with it. Increased hydrodynamic volume decreases renal clearance.

          Homobifunctional Reagents
Homobifunctional reagents are chemical compounds that contain two identical reactive groups. Symmetrical homobifunctional reagents were among the first crosslinkers developed for conjugation of biomolecules. These were typically hydrophobic compounds, and their use – particularly in one-step reactions – often was accompanied with problems such as inactivation and precipitation of the conjugated biomolecules.

Ourproducts are amphiphilic. Therefore, they impart water solubility to molecules to which they are conjugated. Consequently, when used in crosslinking reactions, Bis-dPEG®25-NHS ester will not cause problems such as aggregation, precipitation, and inactivation.
