        • Biofunctionalized fluorescent nanodiamonds are infinitely photostable labels for fluorescent and multiphoton microscopy applicable to cellular tracking and long-term observation of biological processes in the same fields of view in live cell-imaging and fixed cells. Protein, antibody, biologically active organic molecules, and custom functionalizations are possible. We provide nanodiamonds with nitrogen-vacancy (NV) centers with red-to-near-infrared fluorescence, NVN centers with green fluorescence, and novel particles available with a combination of NV and NVN centers within a single particle with visual yellow fluorescence. Fluorescence from NV centers can be magnetically modulated providing a means to eliminate the fluorescent background by image subtraction and to significantly enhance the signal-to-noise ratio. This property can be very useful in future biosensing applications, for example in clinical diagnostics such as lateral flow assays (LFA). Relevant applications also include super-resolution microscopy, multiphoton microscopy, time-gated microscopy and correlative microscopy.

          Background

          The fluorescence properties of fluorescent nanodiamonds (FNDs) are caused by the presence of optically active defects, primarily nitrogen vacancy (NV) centers, on the surface of diamond crystals. The NV center is composed of a carbon vacancy site to one substitutional nitrogen atom at its adjacent site. There are two charge states of NV: one is neutral NV and the other is NV-. NV centers consisting of both neutral and negative charges in diamond show similar photo-physical properties like red fluorescence and bio-imaging properties. There are other types of nitrogen vacancy centres like H3 centre and N3 centre responsible for green and blue fluorescence, respectively. H3 centre comprises of a vacancy adjacent to two nitrogen atoms and N3 centre comprise of a vacancy that is surrounded by three nitrogen atoms. Due to the robust structure of diamond lattice, these defect centres have stable photoluminescence and no photobleaching or photoblinking is observed.
          FNDs have emerged as a powerful tool for analyzing and tracking various biological systems, such as drug delivery, cell tracking, bioimaging, and biolabeling. As fluorescent markers, FNDs possess desirable properties such as high spatial resolution, high molar absorptivity, absence of photobleaching and photoblinking, tunable surface chemistry, inherent biocompatibility, and low toxicity. In bio-imaging, FNDs are used for cell tracking and labeling, showing long-term tracking capabilities without significant exocytosis. FND labeling can be applied to track the regeneration and engraftment of transplanted stem cells. Super-resolution imaging techniques such as Stimulated Emission Depletion Microscopy (STED) enable high-resolution imaging of FNDs in cells. In addition, FNDs are potential tools for temperature sensing at nanoscale level in biological environments. To meet some demanding bioimaging applications, such as advanced protein labeling techniques, the particle size of FNDs must be reduced to 10 nm.
          With the continuous development of nanotechnology, fluorescent nanodiamond is expected to revolutionize biotechnology and promote the progress of medical diagnostics, therapeutics, and other fields.
