        • The Anti-Tau [pS199] antibody (Ref. 4BDX-1502S) recognizes the phosphorylated serine 199 of Tau proteins. This antibody is suitable for Western Blot, for Immunohistochemistry and Immunofluorescence.

          This polyserum was obtained by immunizing a rabbit with a 12 amino acid peptide containing the phosphorylated serine 199 (SGYSSPGSPGT). This product is determined by its ability to recognise human Tau phosphorylated at serine 199.

          Background

          The neurofibrillary degeneration is characterized by an accumulation of fibrils made of hyper and abnormally phosphorylated Tau proteins. The Tau phosphorylation at serine 199 is observed at early stage during the process of neurodegeneration.

          Structurally, the serine 199 in Tau is phosphorylated by the following kinases: CDK5 (cyclin dependent kinase 5), CK2 (casein kinase 2), PKA (protein kinase A), GSK3 (glycogen synthase kinase 3), ERK/MAPK (mitogen activated kinases) and MARK (Microtubule-associated protein/microtubule affinity-regulating kinase).

          The Anti-Tau [pS199] antibody (Ref. 4BDX-1502) detects specifically Tau hyperphosphorylation at serine 199 at early steps of neurofibrillary degeneration in human brain tissue or in animal models of tauopathies.

          Tau protein phosphorylation at serine 199 is then an early marker of neurofibrillary degeneration in Alzheimer’s patients (Maurage et al. 2003).

          Phosphorylated serine 199 epitope is also found in other tauopathies such as progressive supranuclear palsy (Wray et al., 2008). Interestingly, the neurons of young individuals were also shown to express the serine 199 phosphorylated tau-epitope as published in an article of Maurage et al. 2001, using the antibody (Ref. 4BDX-1502) in the paper entitled “A paradoxical expression of serine 199 phosphorylated tau-epitope is found in the hippocampus of young individual”.
