    • Overview
      • This personalized peptide immobilization kit is designed to work with peptides containing amino functional groups. All natural peptides have an N-terminal amino group that can react with carboxylic acid groups. If you have Lysine amino acids, the ε-amino group of lysine may also react.

If you prefer a site specific single-labeling strategy, please check your peptide sequence to make sure only one amino groups present in the peptide. N-terminal amino group can be acetylated during the SPPS to make it inert. You can also add a Lys group in the peptide to position your peptide on the agarose beads if you don't have any amino group.

This kit provides agarose beads derivatized with carboxylic acid functional groups (5-20 μmol COOH per mL of resin). Using the kit components, customers immobilize their amino containing peptide onto SepSphere™ agarose beads in one step following the standard protocol.

        • Key Features of this Immobilization Kit:
        Minimum requirement for immobilization set-up
All reagents, buffers, and plasticwares are included
        Modestly activated carboxylic acid agarose
One step immobilization with very mild reaction conditions
Easy preparation with air push mechanism for washing: less than 1 h hands-on time
Stable linkage (no leakage of the peptides from the resin under acid or base condition)

        • Specification of the Solid Support
        Functional groups: carboxylic acid (COOH)
Matrix: 4% beaded agarose (source: GE Life Sciences, Sepharose® 4B)
        Particle size: 45-165 um
        Mean bead size: 90 um
Ligand concentration: 5-20 μmol COOH per mL agarose

        • Specification of the Immobilized Product
Immobilization chemistry: immobilized via stable amide bond
        Chemical stability: stable
        pH Stability: stable

        • Requirement for peptide
Have at least one amino functional group in the peptide that can react with carboxylic acid. Note: multiple amino groups in a peptide can result in a mixture of immobilized products at various positions
▫ N-terminal amino group if it is not acetylated or blocked
        ▫ ε-amino group of lysine
        HPLC purified and lyophilized
Preferably >90% pure by C18 HPLC
        Total amount: 1-20 mg
