The ExoStar Extracellular Vesicle Analysis System is an absolute quantitative analysis platform for single vesicle subpopulations. The principle is to incubate the extracellular vesicles (EVs) to be tested with antibody-coupled nucleic acid complexes. Following the removal of nonspecifically bound antibody-nucleic acid complexes via ultrafiltration tube washing,single vesicle PCR reaction system prepared by extracellular vesicle analyzer. After the PCR reaction, the fluorescence signal of each microdroplet is recognized and read by the extracellular vesicle analyzer, and the subpopulations of extracellular vesicles are identified and quantified according to the fluorescence signal and Poisson distribution principle.

Workflow
· Generation of reaction mixtures
Incubated and washed EVs are mixed with Extracellular Vesicle Assay Kit.
· Droplet generation
Encapsulation of individual EVs are achieved by generating microdroplets of pre-treated EVs and nucleic acid signal amplification systems on a microfluidic chip via a droplet generator.
· Extracellular vesicle PCR amplification
Transfer to a regular PCR instrument for thermal cycling reaction to amplify antibody-coupled nucleic acid signals.
· Fluorescence signal reading and analysis
The fluorescence signal of each microdroplet is identified and read by the droplet analyzer, and the absolute quantification of single vesicle subpopulation is realized based on the fluorescence signal and Poisson distribution principle.
