nexttec™ 1 -Step is the simplest and least time-consuming DNA purification system, containing all reagents required for lysis and subsequent DNA purification. With nexttec™ DNA isolation systems, DNA is purified in four minutes by a single centrifugation step after cell lysis, which is five times faster than purification using other DNA separation systems.
The nexttec™ 1-Step technology is a reversal of known and common DNA purification systems such as those based on silica membrane and magnetic beads. In contrast to traditional purification methods that retain DNA, the nexttec™ proprietary sorbent retains proteins, detergents and low molecular weight compounds but allows pure DNA to pass through. The nexttec™ system ensures a high DNA recovery rate through avoiding unnecessary pipetting and washing steps.
nexttec™ 1 -Step kits are available in two forms, cleanColumns and cleanPlates96, for isolating pure DNA from a wide range of sample materials such as tissue, cells, plants, blood, plasmid DNA, and bacteria. Trace DNA samples are defined as any sample below recommended thresholds at any stage of the analysis from sample detection to profile interpretation. Trace DNA analysis has become an integral part of a forensic work and an important tool in investigations.
In some cases, touch DNA forensic samples are treated as trace DNA samples based on the potentially low template content. Small amounts of DNA fragments are present on the surface of the skin, and it is speculated that these fragments may be continually shed from the keratinized layers of the skin. Additionally, there may be DNA fragments in sweat. Thus, DNA profiles can be obtained by analyzing the trace DNA left on touched objects.
Based on nexttec™ 1-Step DNA isolation technology, Fingerprint DNA Finder (FDF) is a simple and rapid DNA extraction kit that only takes 30 minutes to digest sample materials and 4 minutes to purify DNA for touch DNA analysis. Using FDF, DNA can be extracted from fingerprints (epidermal cells and cell-free DNA) very simply, without more PCR cycles, which usually increase the imbalance of heterozygotes and are associated with an increase in stutter peak area.
