        • DENARASE® is a genetically engineered endonuclease from Serratia marcescens being able to cleave all forms of DNA and RNA very efficiently. It is commonly used in biotechnology for the quantitative removal of nucleic acids and for viscosity reduction. In the pharmaceutical industry, the endonuclease is often used to remove excess of nucleic acids in production processes of biopharmaceuticals and vaccines.
          c-LEcta has developed a patent-protected production technology for this enzyme based on the recombinant expression in a Bacillus sp. production host. The production process combines high product yields with the advantages of an endotoxin-free production strain. DENARASE® has a purity of ≥ 99 % and is manufactured in full compliance with GMP requirements using only raw materials of non-animal origin and without the use of antibiotics.

          Enzyme Characteristics
          DENARASE is an enzyme consisting of two subunits with a calculated molecular weight of 27 kDa each. The enzyme hydrolyzes phosphodiester bonds between nucleotides and leaves molecules with a length of mainly 2-5 bases with a 5'-monophosphate end.
          DENARASE hydrolyzes both DNA and RNA in various forms like e.g. single strand, double strands, circular and circular supercoiled DNA.
          The enzyme shows high activity under a broad range of circumstances which makes them very attractive for use in Biotech processes.

          The products DENARASE, 1 MU and DENARASE, 5 MU (Article Number: 20804-1M, 20804-5M) are for routine applications (produced and filled under GMP).
          The products DENARASE, 100 kU and DENARASE, 500 kU (Article Number: 20804-100k, 20804-500k) can be used for laboratory test applications (produced under GMP and filled under ISO 9001).

        • Application Description
          DNA can cause high viscosity in bioprocesses and often interacts with the target product, resulting in lower recovery rates. DENARASE reduces the DNA level at an early stage of the process and improves recovery rates. Successful applications of DENARASE include the purification of vaccines, proteins expressed in inclusion bodies and monoclonal antibodies.
          • Biotech products must be free of host cell DNA larger than 100 bases. DENARASE leaves DNA chains of mainly 2-5 bases and can be used as DNA removal technology.
          • DNA can stick to the target molecule such as monoclonal antibodies and viruses. Coverage of binding groups can lead to lower recovery rates for chromatography steps.
          • High DNA concentrations cause high viscosities which makes filtration and chromatography difficult. DENARASE helps to reduce the viscosity and to improve the process economics.
          • DENARASE can prevent cell clumping in bioreactors.
